چکیده

on the capacity of tissue elements to strain metachromatically. Surprising as it may seem, adequately fixed and dehydrated tissues withstand five minutes' immersion in concentrated sulphuric acid without charring or showing any severe disturbance of their structure. Subsequent staining with a metachromatic dye reveals that a greatly increased range of tissue elements shows a metachromatic affinity, and this staining technique offers a new and useful polychrome method which may well have some degree of histochemical specificity. The technique which has been developed involves immersion of sections in concentrated sulphuric acid, followed by staining with a dilute solution of a metachromatic dye. The method is simple, quick, and reliable.

Mast Cells in Experimental Rat Sarcomas*

The mast cells described by Ehrlich (6) are characterized by granules showing a so-called true metachromasia. Using as a basis Lison's statement (22) that this color reaction is typical of macromolecular sulphuric ester compounds and Jorpes' (17) demonstration that heparin is a mucoitin ester sulphuric acid showing a distinct metachromasia, Holmgren and Wilander (11) proved the mast cell granul...

Histochemical Observations on the Cortical Region of the Oocytes of

The cortical cytoplasm of the developing oocyte of Rana pipiens has been investigated by cytochemical means. Particular emphasis is placed on the nature and distribution of polysaccharide protein complexes. With toluidine blue O, both living and fixed mature oocytes were found to possess a distinct metachromatic region just under the developing vitelline membrane. Younger oocytes, at the previt...

A colorimetric method for the standardization of heparin preparations.

IT is a fact well known to histologists that certain basic dyes undergo a change of tint when taken up by particular tissue elements, especially cartilage, mucus and the granules of the so-called 'mast cells'. The chemistry of this phenomenon, named 'metachromasia' by Ehrlich, has been studied in some detail by Lison [1935], This investigator concluded that the change of colour in the dye is du...

Diffusion measurement in ferrous infused gel dosimeters

Background: The compositions of Ferrous sulphate, Agarose and Xylenol orange dye (FAX) and Ferrous sulphate, Gelatin and Xylenol orange dye (FGX) in solution of distilled water and sulphuric acid are two tissue-equivalent gel dosimeters. Ionizing radiation causes oxidation of Fe2+ ion to Fe3+ ions which diffuse through the gel matrix and blur the image of absorbed dose over a period of hours af...

Metachromasia observed in the living eggs of Arbacia punctulata and its cytochemical analysis.

I T has long been known that metachromasia (a change that occurs in the absorption spectrum of certain basic dyes when placed under certain conditions) can be used as a valuable, though not specific, cytochemical method. The physical as well as the chemical conditions promoting metachromasia or affecting its intensity have been studied in vitro on microscopic sections of fixed tissues and, some...
